Adhesion of chicken hepatocytes to polyacrylamide gels derivatized with N-acetylglucosamine.

Complex carbohydrates on the surfaces of eukaryotic cells are thought to participate in a wide variety of cell-cell interactions. A model system has therefore been developed to study these processes. In the present experiments, the ability of chicken hepatocytes to recognize and adhere to sugars covalently linked to polyacrylamide gels was investigated. The gels were snythesized by two methods....

Selective binding of N-acetylglucosamine to the chicken hepatic lectin.

Among Ca2+-dependent (C-type) animal lectins, the chicken hepatic lectin (CHL) is unique in displaying almost complete selectivity for N-acetylglucosamine over other monosaccharide ligands. The crystal structures of the carbohydrate-recognition domain (CRD) from serum mannose-binding protein (MBP) and of a complex between the CRD from liver MBP and the methyl glycoside of N-acetylglucosamine we...

Adhesion of Dictyostelium discoideum cells to carbohydrates immobilized in polyacrylamide gels. I. Evidence for three sugar-specific cell surface receptors.

Dictyostelium discoideum cells appear to be able to recognize particular carbohydrate prosthetic groups at different stages in their life cycle. We therefore used our previously developed model system (consisting of polyacrylamide gels containing putative ligands covalently linked to the polymer) to determine the receptors on these cells capable of recognizing carbohydrates. D. discoideum cells...

Formation and Structure of Polyacrylamide Gels

Acrylamide and N,N-methylenebis(acry1amide) (AAm-Bis) copolymerization has been investigated in water at a monomer concentration of 1.8 w/v %. Conversion of monomer and pendant vinyl groups was measured as a function of the reaction time up to the onset of macrogelation. Experimental results indicate that 80% of pendant vinyl groups are consumed by cyclization reactions. When the monomer concen...

Demonstration of rhodanese activity in polyacrylamide gels.

Rhodanese activity from crude extracts of Thiobacillus sp. strain IV-85 was demonstrated in polyacrylamide gels after incubation in the reaction mixture by staining with dichloroindophenol in the presence of methylphenazonium methosulfate. The sensitivity of the staining system was found to be 8 x 10 moles of sulfite.
